    • Regulation of the neutrophil response by T-cell derived cytokines impacts upon Staphylococcus aureus infection outcomes (WT086515MA)
  • Summary
    • This proposal investigates how T-cells activated in the context of acute S.aureus infection can modulate the local neutrophil response. A clinically relevant murine model of S. aureus wound infection will be employed in conjunction with knockout mice to establish if specific T-cell phenotypes present at sites of S.aureus infection and the cytokines they produce can control neutrophil trafficking, activation and survival. The major focus of this grant is the neutrophil response during S.aureus infection. In particular it will investigate the controversial concept that S.aureus can exploit the neutrophil response and survive intra-cellular within the neutrophil. The conditions that favor this phenomena and the mechanism involved will be determined e.g. rate of neutrophil apoptosis/necrosis, phagocytic activity, and authophagy. In particular we will investigate if T-cell derived cytokines can modulate these phenomena.

  • Summary
    • This proposal will investigate the immunomodulatory properties of a capsular polysaccharide molecule expressed by the organism Streptococcus pneumoniae, which is commonly found to colonise the nasopharynx. This molecule has been shown to possess a unique chemical structure that confers upon it the ability to activate immunomodulatroy T-cells to produce the anti-inflammatory cytokine Interleukin-10. We will now investigate the ability of this polysaccharide to induce IL-10 production and protect against experimental asthma.
