Research Expertise

Description

My research is focused on molecular aspects of sarcoma and targeted therapeutics in cancer. In addition, I have an interest in cancer immunology. I have an MSc in the epigenetics of malignant disease and this also informs my research output. As a sample example of my research output, my publication in The Journal of Adolescent and Young Adult Oncology run from CHOC Hospital in Los Angelus provides a novel perspective on the role of the innate immune system in the pathogenesis of osteosarcoma. My publications have been cited in leading journals including Proceedings of the National Academy of Sciences, Cancer Cell, Journal of Clinical Oncology, Elife, Cancer Research, Nature Communications, Journal of Biological Chemistry, Nature Reviews Cancer, Development, JCO Precision Oncology, Oncogene, and iScience (Cell Press). Projects

  • Title
    • Transcriptome analysis, proteomic profiling, and computational analysis of a series of melanomas to discover potential antigen candidates to be targeted by chimeric antigen receptor genetically engineered T cells
  • Summary
    • Treatment of melanoma with checkpoint inhibitors has been one of the greatest recent therapeutic advances in oncology. CAR-T cells have shown remarkable efficacy in B cell malignancies. The inability to identify a suitable antigenic target has hindered the theoretical promise of CAR T cell treatment in these immunogenic cancers. It is intended to prospectively compile a clinical series of melanoma samples from St. James Hospital. RNA sequencing of these in melanoma will be undertaken to establish the transcriptome. 2D-SDS PAGE and mass spectroscopy to determine the proteome. Computational analysis will be applied to elucidate the protein function to select potential tumor-associated antigen candidates. Murine melanoma models utilized for adoptive T cell transfer of the constructed gene for chimeric antigen receptor to establish pre-clinical efficacy. • Title
    • Elucidation of the mechanisms of action underlying immune checkpoint inhibitor therapy to improve treatment response
  • Summary
    • To examine the effects of checkpoint inhibitors on a wide range of effector lymphocyte subsets. There is a particular focus on unconventional T cell subsets with potent anti-tumor effector functions, for example, gamma/delta T cells, mucosal-associated invariant T (MAIT) cells, and invariant natural killer cell T (iNKT) cells Submitted for SFI-AICRI Grant.
