The main thrust of my research work has been the study of membrane processes in Trypanosomes. SIGNIFICANT MILESTONES IN THIS WORK: A. First studies of amino acid transport in trypanosomes, (refs 1-2, 5-8, 10 & 14) B. Development of the current method used for the commercial production of [32P]ATP (ref 9) C. First production of isolated plasma membranes in trypanosomes (ref 12) D. Development of a mild method for selectively and reversibly permeablizing plasma membranes to small molecules while leaving other cellular membranes unaffected, (ref 16) E. First series of studies characterizing the adenylate cyclase in bloodstream form trypanosomal plasma membranes and discovery of the regulation of this enzyme by Ca2+, (ref 11, 16-17, 20 & 38) F. First study of glycoprotein biosythesis in trypanosomes, (ref 15) G. Discovery of the phosphodiester linkage of the variant surface glycoprotein to the lipid anchor and its cleavage during release from the cell surface, (ref 18-19, 21, 23 & 25) H. First series of studies of the bioenergetics of bloodstream form trypanosomes, (refs 31,33,35, 44-45) I. Discovery and characterization of invariant surface glycoproteins on the surface of bloodstream form trypanosomes and demonstration of their life cycle specificity, (ref 40, 43 & 47) J. Discovery of the ability of bloodstream stage trypanosomes to defend themselves against antibody without cell damage, (ref 41 & 49) K. First 3-D structure of the C-terminal folding domain of the variant surface glycoprotein (VSG) in trypanosomes (ref. 48) L. Development of a new continuous, pre-steady state, single turn-over fluorescence assay for the endocytotic-exocytotic cycle and its use to characterize the endocytosis of surface immune complexes and nutritional factors, (refs 49-50) M. Development of Specific Fluorescent Phospholipid Probes for the Plasma Membrane of all Eukaryotic Cells (Patent Pending, European Patent Office, submitted 2004) N.
