Biography

I was awarded a PhD in neuroscience from the Department of Anatomy and Neuroscience, University College Cork in 2005. I subsequently conducted postdoctoral research into genetic brain disorders at the Department of Pathology and Cell Biology, Columbia University, New York. From 2008 until 2011, I undertook postdoctoral research fellowship positions focusing on HIV-1 pathogenesis and cardiovascular diseases at the Centre for Research into Infectious Diseases and the Conway Institute of Biomolecular and Biomedical Research, University College Dublin. In 2011, I was appointed to the position of Lecturer/Principle Investigator in the Department of Anatomy and Neuroscience, University College Cork. In 2013, I was appointed to the position of Assistant Professor in the Department of Anatomy, Trinity Biomedical Sciences Institute, TCD. In August 2016, I was confirmed in post as Assistant Professor, TCD.

Research Expertise

Description

My main research focuses on the lineages, functions and restorative potential of neural precursor cells. Radial glial cells are elongated neural progenitor cells present in the CNS during development that represent a major progenitor pool during development and can give rise to neurogenic niches of the adult. I have shown that radial glial cells are essential for correct white matter formation in the spinal cord forming glial scaffolds that guide growing axons to their synaptic destinations. A primary research goal is gaining insights into the potentials of radial glial cells, and their axon scaffolding properties, in restoring connectivity in the spinal cord using an in vitro injury model. Three dimensional imaging, using new methods such as 3DISCO, of these scaffolds should enhance our understanding of the axon-glial interactions occurring within them. We aim to rigorously characterise these scaffolds and the biomarkers they express to shed light on their potentials in restoring tract formation in injury scenarios. As such, biomarkers such as proteoglycans are also showing much potential in identifying subsets of neural cells that are responsible for mediating tract formation and for determining the fates of multipotent stem cells. Glycans regulate the activity of membrane receptors and mediate cell-cell or cell-matrix interactions. LewisX (LeX) is a glycan motif associated with glycoproteins and glycolipids that is present on neural stem and progenitor cells and acts in association with Wnt and notch signalling. We have shown that LeX is present on discrete subpopulations of neuroepithelial and radial glial cells in the developing brain and spinal cord. However, while glycoconjugates are expected to play significant roles in stem cell therapies, the basic functional relevance of LeX glycosylated neuroepithelial cells and radial glial cells is unknown in the developing CNS.My interest in the developmental origins of brain cells is forming the basis for research linking dietary influences, such as the ketone diet (KD), to neural stem cell functions during pregnancy. The KD (low carbohydrates/ high fat) have become popular recently due to their effectiveness for weight loss and in treatment of neurological disorders such as epilepsy. Although the diet is a promising alternative to medication for those suffering from epilepsy, there is limited information about how the diet affects normal brain functioning. Furthermore, there is very little research on how KD during pregnancy may affect the developing brain. This research question becomes increasingly important as women of reproductive age may be on the diet for extended periods of time. Anatomy based research interests include the investigation of the use of Diffusion Weighted-MR Imaging (DW-MRI) for white matter tracts in vivo. Concerns remain about the robustness and anatomical relevance of this DW-MRI in probing more convoluted tracts as in the limbic system. Complex white matter architecture (crossing fibres) and methodological constraints (signal to noise ratio (SNR) and b-values become a significant issue in tract generation affecting the accuracy of the tract formations. In collaboration with investigators at TCD, I am comparing the validity of the rendered limbic white matter tracts using advanced tractography techniques to "gold standard" anatomical cadaveric dissection measurements. The goal is to reveal limbic connectivity in higher detail than has been previously described.Moreover, I am currently supervising MD research aiming to provide greater insights into the spatial relationships between the facial nerve and retromandibular vein in human cadaveric parotid glands. This research is especially significant in the context of parotid surgery. I also conduct education research based on the roles of social media in anatomy education and the insights of radiologists and surgeons on the most efficient modes of anatomy training for professional practice.
