Instrumentation

The Flow Cytometry Facility is equipped with one flow sorter and three analysers:

Dako MoFlo Cell Sorter:

The MoFlo High Performance Cell Sorter is a modular flow cytometer capable of sort speeds up to 70,000 cells or particles per second. With a Coherent Enterprise II 488nm (Blue) and 351nm (UV) water cooled laser, and a solid state 635nm, 25mW (Red) diode laser, three fluorescent channels are picked up from the 488nm (Blue) laser [FL1, FL2, FL3], two from the 351nm (UV) laser [FL4, FL5], and one from the 633nm (Red) laser [FL6]. The cytometer is capable of up to 4 sorts simultaneously into a variety of different receptacles. Also, single cell deposition into most standard 80mm x 120 mm multi-well tray formats (e.g. 96-well plate) and onto standard slides is routine with the Cyclone system.

Dako CyAn ADP:

The CyAn ADP (Advanced Digital Processing) is a benchtop flow cytometer capable of analysis of up to 50,000 cells per second. Our CyAn ADP LX 7 Colour model is equipped with 488nm (Blue) and 633nm (Red) solid state lasers. The nine parameters available include FSC, SSC, and 7 fluorescent channels (FL1-FL7). This model has five fluorescent channels available from blue laser and two from red laser excitation. This cytometer is hence ideal for up to seven colour flow analysis. The CyAn ADP is operated through the Summit Software. Here, flexible management tools allow you to define samples, workspaces, and monitor instrument status in parallel. With features such as auto-compensation, post-acquisition compensation, and voltage and compensation settings saved with the FCS file etc. instrument set-up, acquisition and analysis are straightforward.

BD FACSCalibur:

The Becton Dickinson FACSCalibur is a three colour benchtop flow cytometer system, well suited to routine analysis on cell samples with up to three fluorescent labels. The single 15mW 488nm (Blue), air-cooled argon-ion laser allows the two scatter parameters (FSC and SSC) and three fluorescent channels to be detected (FL1-FL3). Our FACSCaliburs are single-sample load cytometers, with 3 variable acquisition rates. The cytometers are run using Apple Macintosh computers running CellQuest software on OS 9.0.
